Activity of fungal phytases stored in two ways in response to the period of storage at room temperature Ciência Rural
Naves,Luciana de Paula; Corrêa,Angelita Duarte; Bertechini,Antônio Gilberto; Gomide,Elisangela Minati; Abreu,Celeste Maria Patto de.
Two distinct experiments were conducted simultaneously with phytases of Aspergillus oryzae and A. niger for determining enzyme activity in response to storage period (up to 180 days at room temperature - RT). In the first experiment, enzymes were stored as they were acquired (pure form) and the activity was measured periodically during 180 days of storage at RT. In the second experiment, the phytases were incorporated to a supplement containing vitamins, minerals, and amino acids; and then this supplement was stored at RT up to 180 days, so that every 30 days of storage was collected one aliquot from each replicate for determining enzymatic activity. In conclusion, the phytase activity is affected by storage duration. To ensure 80% of the initial activity,...

Animal manure phosphorus characterization by sequential chemical fractionation, release kinetics and 31P-NMR analysis Rev. Bras. Ciênc. Solo
Tiecher,Tales; Zafar,Mohsin; Mallmann,Fábio Joel Kochem; Bortoluzzi,Edson Campanhola; Bender,Marcos Antonio; Ciotti,Lucas Henrique; Santos,Danilo Rheinheimer dos.
Phosphate release kinetics from manures are of global interest because sustainable plant nutrition with phosphate will be a major concern in the future. Although information on the bioavailability and chemical composition of P present in manure used as fertilizer are important to understand its dynamics in the soil, such studies are still scarce. Therefore, P extraction was evaluated in this study by sequential chemical fractionation, desorption with anion-cation exchange resin and 31P nuclear magnetic resonance (31P-NMR) spectroscopy to assess the P forms in three different dry manure types (i.e. poultry, cattle and swine manure). All three methods showed that the P forms in poultry, cattle and swine dry manures are mostly inorganic and highly...

Bacterial phytase: potential application, in vivo function and regulation of its synthesis BJM
Konietzny,Ursula; Greiner,Ralf.
The stepwise release of phosphate from phytate, the major storage form of phosphate in plant seeds and pollen, is initiated by a class of enzymes that have been collectively called phytases. The classification is solely due to the in vitro capability of these enzymes to accept phytate as a substrate. Phytases have been studied intensively in recent years because of the great interest in such enzymes for reducing phytate content in animal feed and food for human consumption. They have a wide distribution in plants, microorganisms, and in some animal tissues. Due to several biological characteristics, such as substrate specificity, resistance to proteolysis and catalytic efficiency, bacterial phytases have considerable potential in commercial applications....

Bioavailability and poultry fecal excretion of phosphorus from soybean-based diets supplemented with phytase R. Bras. Zootec.
Martins,Bianca Almeida Brandão; Borgatti,Laura Maria de Oliveira; Souza,Luiz Waldemar de Oliveira; Robassini,Simi Luiza Durante Aflalo; Albuquerque,Ricardo de.
Five hundred and four 1-day-old Cobb broiler chickens were randomly allotted to 18 treatments and fed a semi-purified basal diet (4 g/kg total phosphorus) supplemented with 2 levels of supplemental P (0.5 or 1 g/kg) during 21 days to evaluate performance and bone mineralization; to assess the relative bioavailability of P (RBP) in wheat meal (WM) and roasted (RS) or extruded soybean meal (ES), with or without phytase (0 or 750 ftu/kg diet), using dicalcium phosphate (DC) as standard; and to estimate phosphorus excretion. Weight gain (WG), feed intake (FI), feed to gain ratio (FG), and tibia ash weigh (AW) were assessed. For each source, the RBP was calculated by the slope ratio method, when tibia ash weight was regressed on P ingested. Ingested and fecal P...

Effect of ph and temperature on the activity of phytase products used in broiler nutrition Rev. Bras. Ciênc. Avic.
Naves,L de P; Corrêa,AD; Bertechini,AG; Gomide,EM; Santos,CD dos.
The activity of three commercial microbial phytase (Aspergillus oryzae, A. niger, and Saccharomyces cerevisae) products used in broiler nutrition was determined at different pH (2.0 to 9.0) and temperature (20 to 90°C) values. Enzymatic activity was determined according to the reaction of the phytase with its substrate (sodium phytate), in four replicates, and was expressed in units of phytase activity (FTU). A. oryzae phytase exhibited optimal activity at pH 4.0 and 40°C, but its absolute activity was the lowest of the three phytases evaluated. A. niger phytase exhibited maximal activity close to pH 5.0 and 45ºC, whereas S. cerevisae phytase presented its highest activity at pH close to 4.5 and temperatures ranging between 50 and 60°C. It was concluded...

MEDICI,LEONARDO O.; GONÇALVES,FABÍOLA V.; FONSECA,MARCOS PAULO S. DA; GAZIOLA,SALETE A.; SCHMIDT,DAIANA; AZEVEDO,RICARDO A.; PIMENTEL,CARLOS.
ABSTRACT In this study, we are presenting recommendations to the best agricultural use as well as for plant breeding of three millet cultivars namely ENA1 and ENA2, which have African origin, and BRS1501 originally from India. These cultivars were evaluated for growth, yield and grain quality traits. The morphological traits evaluated in this study indicated that the African genotypes ENA1 and ENA2 are better than the Indian genotype BRS1501 for no-till farming or to produce forage with 15% of crude protein at flowering and at harvest to produce stover (around 7% of crude protein content) for livestock feeding. The BRS1501 cultivar exhibited the highest values for total crude protein, albumins and prolamins, phytate and mineral contents in grains. ENA1 and...

Heat-resistant bacterial phytase in broiler pelleted diets Rev. Bras. Ciênc. Avic.
Carlos,TC de F; Barbosa,LCGS; Shiroma,NN; Dari,RL; Baolin,G; Yongcheng,W; Araújo,CS da S; Araújo,LF.
The objective of the study was to evaluate the effects of a heat-resistant bacterial phytase added to pelleted diets on mineral digestibility, live performance, carcass traits, and bone quality of broilers. Three treatments were evaluated: Positive control; negative control, with 0.10 points reduction in calcium level and 0.15 points reduction in available phosphorus level; and negative control + phytase at 500 FTU/kg. Mineral digestibility and bone quality results demonstrated that the evaluated phytase resisted pelleting as it increased the utilization of the minerals present in the diet.

Increasing levels of phytase in diets formulated with reduced available phosphorus content supplied to male and female broilers Ciência Rural
Naves,Luciana de Paula; Rodrigues,Paulo Borges; Teixeira,Levy do Vale; Bertechini,Antônio Gilberto; Alvarenga,Renata Ribeiro; Bernardino,Verônica Maria Pereira; Santos,Luziane Moreira dos; Natividade,Yuri Pereira Efrem.
The reduction of the available phosphorus (avP) content in the broiler diet must be followed by the supplementation of adequate phytase. One experiment was conducted with broilers from 15 to 28 days of age to determine the level of necessary phytase when the mashed diet is formulated with 0.213% of avP to enable the results of performance, tibia ash content, and calcium (Ca) utilization similar to those determined for birds fed with diet formulated to meet their nutritional requirements. Thus, 120 broilers were distributed in (4+1) x2 factorial arrangement corresponding to four deficient diets in avP (0.213%) supplemented with phytase (0; 750; 1,500 or 2,250FTU kg-1) plus one positive control diet without phytase (0.426% of avP), supplied to male and...

Isolation, morphological and molecular characterization of phytate-hydrolysing fungi by 18S rDNA sequence analysis BJM
Gontia-Mishra,Iti; Deshmukh,Dhanshree; Tripathi,Niraj; Bardiya-Bhurat,Khushboo; Tantwai,Keerti; Tiwari,Sharad.
Phytate is the primary storage form of phosphate in plants. Monogastric animals like poultry, pigs and fishes have very low or no phytase activities in their digestive tracts therefore, are incapable to efficiently utilize phytate phosphorus from the feed. Phytase from microbial sources are supplemented to feedstuff of these to increase the uptake of phytate phosphorus. In the present work efforts were made to isolate and characterize proficient phytase producing fungi from soil. Phytase producing fungi were isolated using phytate specific medium. Fungal isolates were selected according to their higher phytase activities. These isolates were further characterized and identified by morphological and microscopic analysis and confirmed by amplification of 18S...

Phytase enzyme in diets containing defatted rice bran for growing swine Scientia Agricola
Moreira,José Aparecido; Vitti,Dorinha Miriam Silber Schmidt; Trindade Neto,Messias Alves da; Lopes,João Batista.
Organic phosphorus is poorly utilized by monogastric animals because they lack phytase, the enzyme that cleaves the ortho-phosphate groups from the phytate molecule. Diets fed to pigs are supplemented with inorganic P, and this can increase environmental pollution and diet costs. Sixty mixed sex, half-breed pigs, were used to evaluate the effect of increasing dietary levels of phytase (253, 759, 1265 and 1748 PU kg-1 feed) on animal performance as compared to a control without phytase but supplemented with dicalcium phosphate. Enzyme levels did not affect daily feed intake, food conversion, average daily weight gain, plasma P and Ca, calcium and phosphorus in bone ash, and the calcium/phosphorus ratio in the plasma and bones. A quadratic relationship...

Purification and characterisation of an extracellular phytase from Aspergillus niger 11T53A9 BJM
Greiner,Ralf; Silva,Lucineia Gomes da; Couri,Sonia.
An extracellular phytase from Aspergillus niger 11T53A9 was purified about 51-fold to apparent homogeneity with a recovery of 20.3% referred to the phytase activity in the crude extract. Purification was achieved by ammonium sulphate precipitation, ion chromataography and gel filtration. The purified enzyme behaved as a monomeric protein with a molecular mass of about 85 kDa and exhibited maximal phytate-degrading activity at pH 5.0. Optimum temperature for the degradation of phytate was 55°C. The kinetic parameters for the hydrolysis of sodium phytate were determined to be K M = 54 µmol l-1 and k cat = 190 sec-1 at pH 5.0 and 37°C. The purified enzyme was rather specific for phytate dephosphorylation. It was shown that the phytase preferably...
